Lorlatinib is used in the treatment of Non-small cell lung cancer.

How Lorlatinib works
Lorlatinib is an anti-cancer medication. It works by blocking the action of the abnormal protein that signals cancer cells to multiply. This helps to stop or slow the spread of cancer cells.
Common side effects of Lorlatinib
Anemia (low number of red blood cells) is the major side effect of this drug.
How should I take Lorlatinib ?
You should take Lorlatinib exactly as directed by your doctor. It should be started by doctors who have experience in administering cancer medicines. Take it once a day with or without food. Do not drink grapefruit juice or eat grapefruit during your treatment with Lorlatinib , as the chances of side effects may be more.
What should I do if I forget a dose?
If you miss a dose of Lorlatinib by less than 12 hours, take the missed dose as soon as you remember it and then take the next dose at the scheduled time. However, if you miss a dose by more than 12 hours, skip the missed dose and continue your regular dosing schedule. Do not take a double dose to make up for a missed one.
In case of emergency/overdose
In case of overdose, call the poison control helpline. If the victim has collapsed, had a seizure, has trouble breathing, or can't be awakened, immediately call a doctor.

Lorlatinib is an ATP-competitive inhibitor of the receptor tyrosine kinases anaplastic lymphoma kinase (ALK) and C-ros oncogene 1 (Ros1) with potential antineoplastic action that is accessible orally. Lorlatinib binds to and inhibits both ALK and ROS1 kinases after treatment. Inhibition of the kinase causes ALK and ROS1-mediated signalling to be disrupted, which limits tumour cell proliferation in ALK- and ROS1-overexpressing tumour cells. PF-06463922 also has the ability to pass the blood-brain barrier.
